Background and context

How the trial was designed

ZUPREME-1 is registered as a randomised, double-blind, placebo-controlled, dose-finding Phase 2 trial. It compares five doses of once-weekly petrelintide with placebo in 493 adults at sites in the United States, Poland and Romania. Treatment lasted 42 weeks, followed by a nine-week safety period. Adults needed a BMI of at least 30, or at least 27 with hypertension or dyslipidemia, and people with diabetes were excluded.

Reading the announced figure against the registry

The registry lists percent change in body weight at week 28 as the primary endpoint. The week 42 weight change quoted in the release is a registered secondary measure. The registry names Zealand Pharma as the sponsor, while the release cited here is from Roche. It lists the study as completed, with no results posted, and primary completion in September 2025.

Trial record: ClinicalTrials.gov NCT06662539, read on 2026-10-15.
